ABSTRACT

Innate Immune Recognition and Signaling in Drosophila and Humans

Innate immunity is highly conserved throughout the animal kingdom. A major issue in the field has been to understand how pathogenic microbes are specifically recognized by the innate immune system without the aid of recombinant antigen receptors (T-cell Receptors and antibodies). In a seminal paper sixteen years ago Charles Janeway formalized a theory of pathogen-associated molecular patterns (PAMPs) that would be common, immutable, conserved components of broad groups of microbes, and pattern recognition receptors (PRRs) that would be expressed by the cells of the innate immune system and would recognize these PAMPs. In the intervening years, the Toll-like Receptors (TLRs) have been found to be one major type of PRR in mammals. Humans express at least 10 TLRs, each of which recognizes different types of PAMPs. For example, TLR4 is required for recognition of LPS (endotoxin) while TLR3 recognizes dsRNA from viruses. The TLRs were discovered based on their homology to Toll, which plays a central role in recognizing and responding to Gram-positive bacterial and fungal infections in the Drosophila. The paradigm in Drosophila is slightly different in that Toll appears to function as a cytokine receptor as opposed to a PRR. In addition to the Toll dependent responses, Drosophila have a second recognition system, known as the Imd pathway, that responds to Gram-negative bacterial infection.

The Drosophila immune response lacks all aspects of adaptive immunity and relies entirely on innate immune responses. The Drosophila immune response relies on a different family of PRRs, the Peptidoglycan Recognition Proteins (PGRPs), to recognize bacterial pathogens, in particular bacterial peptidoglycan. Drosophila encode 13 PGRP genes, some of which have multiple splice isoforms. Upstream of Toll, PGRP-SA and PGRP-SD recognize Gram-positive peptidoglycan and stimulate the production of the Toll ligand Spatzle. The Imd pathway relies on two other PGRPs, PGRP-LC and PGRP-LE, to recognize Gram-negative DAP-type peptidoglycan. I will present our latest findings on the molecular mechanisms of recognition by PGRP-LC and PGPR-LE. In brief, PGRP-LC is a cell surface receptor for DAP-type peptidoglycan. PGRP-LC encodes three distinct proteins by alternative splicing and different splice isoforms are required for recognition of monomeric versus polymeric DAP-type peptidoglycan. In addition, PGRP-LE appears to function as an intracellular receptor for the recognition of monomeric peptidoglycan. The intracellular signaling pathways of the Imd and Toll pathways are highly homologous to thesignaling pathways downstream of the TLRs in mammals. All of these innate immune signaling pathways lead to the activation of NF-?B transcription factors, which are central to regulating innate immune response target genes, including antimicrobial peptide genes (in flies and mammals) and inflammatory cytokines (in mammals). In particular, the Drosophila Toll pathway is homologous to the mammalian MyD88-dependent pathway important in TLR2, 4, and 9 responses, while the Imd pathway is more similar to the MyD88-independent pathway critical in TLR3 and 4 signaling. New discoveries linking the Imd and MyD88-independent pathways will be presented.

性質の異なる材料を組み合わせてそれらの特長を活かそうとする方法は昔から行われてきましたが、それらの複合相の単位はミリやミクロン以上でした。しかし、より高機能・高性能な材料の開発へと研究がシフトするに伴い、複合相の単位がだんだん小さくなってナノレベルあるいはそれ以下になってきています。複合相の単位がナノメーター以下になって分子レベルに近づいてくると、単純な機械的混合では複合化が不可能になります。無機・有機ハイブリッドは、従来の無機材料と有機材料から成る複合材料とは異なり、無機物と有機物を分子レベルで融合させながら構造制御したものです。無機物と有機物を分子レベルで融合するためには、有機成分の特性を損なわないようにセラミックスやガラスなどの無機物を低温で合成する必要があり、化学反応によってセラミックスやガラスを低温形成できるゾル・ゲル法が多用されています。我々は、約十年前からゾル・ゲル法を用いて無機・有機ハイブリッドの合成と機能設計について研究を行ってき、無機材料と有機材料との性質を兼ね備えたり、これらの性質の相乗効果(シナジー効果)を有する新規機能を探索してきました。具体的には種々の無機成分を含むオルガノシロキサン系無機・有機ハイブリッドの合成技術および柔軟性等の機械的性質や分子に対する親和力等の化学的性質などの機能設計を行い、自動車エンジンのピストンリングや光学式センシング材料を検討してきました。

主催者追記:有機・無機ハイブリッドとは無機と有機を分子レベルで融合させた材料で、変形できるセラミックスとして自動車のピストンリング向けに開発されています。歯科材料への応用を考えると現状では強度改善が必要ですが、耐摩擦性が優れており複合比を変えることで機械的性質は改善可能であり、今後期待される新素材です。歯科材料への応用に関するアイデアを御検討ください。

第1回 抄録

血栓の形成には、血管を閉塞することによって出血を止めるという働きがある。止血は生命が存続するために必須の機能であり、血栓形成は重要な生体防御反応の一つと言える。一方、血栓の形成は血液循環を阻害することによって、心筋梗塞、脳梗塞などの血栓症や血栓性疾患を発症させる。血栓性疾患は、成人病あるいは生活習慣病のひとつであり、ガンと並んで現代の死因の主要なものである。従って、その予防と治療がガン克服とともに強く求められている。一方、歯周病も成人病や生活習慣病の側面をもち、最終的には歯牙の脱落に至り、QOL を損ねるため、その治療・予防法の確立は 21 世紀には必須である。歯周病の病因論については、すでに確立されているかのように考えられているが、「全身の一部としての口腔」、あるいは「成人病・生活習慣病」といった観点からの考察がまだまだ不足しているように思われる。

血栓症、歯周病ともに、その病態形成には血液循環の障害が関与しており、その基盤には血栓形成異常あるいは血液凝固異常があると考えられる。最近我々は、血管でつくられる一酸化窒素(NO)が血管内における血栓形成を抑える仕組みを明らかにした。すなわち、NOがその分子ターゲットである NSF の活性を制御することによって、血栓形成を抑えることを示した。さらに、NSFの活性を抑える複合ペプチドを用いて、血栓形成を制御できる可能性を示した。また、歯周病原菌である Porphyromonas gingivalis が産生する酵素 gingipainが血液凝固を亢進し、このことが歯周病の病因に深くかかわっていることを示すとともに、同酵素をターゲットにした新規治療薬の可能性についても検討した。  本フォーラムでは、血栓症・歯周病の病因に深くかかわる血栓形成・血液凝固異常の分子メカニズムについて解説するとともに、それらの分子をターゲットとした21世紀型の新しい治療のパラダイムを紹介する。
